Sulfonyl divalent aryl or heteroaryl hydroxamic acid compounds
A sulfonyl divalent aromatic or heteroaromatic ring hydroxamic acid compound that inter alia inhibits matrix metalloprotease activity is disclosed as are a treatment process that comprises administering a contemplated sulfonyl divalent aromatic or heteroaromatic ring hydroxamic acid compound in a MMP enzyme-inhibiting effective amount to a host having a condition associated with pathological matrix metalloprotease activity.
What is claimed is:
1. A compound corresponding to Formula I: ##STR623##
wherein
y and z are each zero or one and the sum of z+y is one;
the ring structure W is a 6-membered divalent aromatic or heteroaromatic ring;
R.sup.1 is a 5- or 6-membered cyclohydrocarbyl, heterocyclo, aryl, or heteroaryl radical bonded directly to the depicted SO.sub.2 -group and substituted with R.sup.4 ;
R.sup.2 and R.sup.3 are independently hydrido, C.sub.1 -C.sub.4 hydrocarbyl, hydroxyl, or amino; and
R.sup.4 is an optionally substituted radical, wherein the radical is selected from the group consisting of single-ring cyclohydrocarbyl, single-ring aryl, single-ring heteroaryl, phenyl, phenoxy, thiophenoxy, anilino, phenylazo, phenylureido,benzamido, nicotinamido, isonicotinamido, picolinamido, heterocyclo, heterocyclohydrocarbyl, arylheterocyclohydrocarbyl, arylhydrocarbyl, heteroarylhydrocarbyl, heteroarylheterocyclohydrocarbyl, arylhydrocarbyloxyhydrocarbyl, aryloxyhydrocarbyl,hydrocarboylhydrocarbyl, arylhydrocarboylhydrocarbyl, arylcarbonylhydrocarbyl, arylazoaryl, arylhydrazinoaryl, hydrocarbylthiohydrocarbyl, hydrocarbylthioaryl, arylthiohydrocarbyl, heteroarylthiohydrocarbyl, hydrocarbylthioarylhydrocarbyl,arylhydrocarbylthiohydrocarbyl, arylhydrocarbylthioaryl, arylhydrocarbylamino, heteroarylhydrocarbylamino, and heteroarylthio, wherein:

the substituent(s) on the monosubstituted or disubstituted aminohydrocarbyl nitrogen is/are selected from the group consisting of hydrocarbyl, aryl, arylhydrocarbyl, cyclohydrocarbyl, arylhydrocarbyloxycarbonyl, hydrocarbyloxycarbonyl, andhydrocarboyl, or
the substituents on the di-substituted aminohydrocarbyl nitrogen, together with the aminohydrocarbyl nitrogen itself, form a 5- to 8-member heterocyclic or heteroaryl ring.

The compound according to claim 1 wherein R.sup.4 is selected from the group consisting of a phenyl group, a phenoxy group, a thiophenoxy group, an anilino group, a phenylazo group, a phenylureido, a benzamido, a nicotinamido, anisonicotinamido, a picolinamido group, a heterocyclo, heterocyclohydrocarbyl, arylheterocyclohydrocarbyl, arylhydrocarbyl, heteroarylhydrocarbyl, heteroarylheterocyclohydrocarbyl, arylhydrocarbyloxyhydrocarbyl, aryloxyhydrocarbyl,hydrocarboylhydrocarbyl, arylhydrocarboylhydrocarbyl, arylcarbonylhydrocarbyl, arylazoaryl, arylhydrazinoaryl, hydrocarbylthiohydrocarbyl, hydrocarbylthioaryl, arylthiohydrocarbyl, heteroarylthiohydrocarbyl, hydrocarbylthioarylhydrocarbyl,arylhydrocarbylthiohydrocarbyl, arylhydrocarbylthioaryl, aryihydrocarbylamino, heteroarylhydrocarbylamino, and a heteroarylthio group.

The compound according to claim 1 wherein R.sup.4 is a substituted radical, the radical being selected from the group consisting of phenyl, phenoxy, thiophenoxy, anilino, phenylazo, phenylureido, benzamido, nicotinamido, isonicotinamido,picolinamido, heterocyclo, heterocyclohydrocarbyl, arylheterocyclohydrocarbyl, arylhydrocarbyl, heteroarylhydrocarbyl, heteroarylheterocyclohydrocarbyl, arylhydrocarbyloxyhydrocarbyl, aryloxyhydrocarbyl, hydrocarboylhydrocarbyl,arylhydrocarboylhydrocarbyl, arylcarbonylhydrocarbyl, arylazoaryl, arylhydrazinoaryl, hydrocarbylthiohydrocarbyl, hydrocarbylthioaryl, arylthiohydrocarbyl, heteroarylthiohydrocarbyl, hydrocarbylthioarylhydrocarbyl, arylhydrocarbylthiohydrocarbyl,arylhydrocarbylthioaryl, arylhydrocarbylamino, heteroarylhydrocarbylamino, and heteroarylthio, wherein:

the ring structure W is a 6-membered divalent aromatic or heteroaromatic ring;
R.sup.1 is a 5- or 6-membered cyclohydrocarbyl, heterocyclo, aryl, or heteroaryl radical bonded directly to the depicted SO.sub.2 -group and substituted at its own 4-position when a 6-membered ring or at its own 3- or 4-position when a 5-memberedring with a substituent, R.sup.4, selected from the group consisting of one other single-ringed cyclohydrocarbyl group, a single-ringed heterocyclo group, a single-ringed aryl group, a single-ringed heteroaryl group, a phenoxy group, a thiophenoxy group,a 4-thiopyridyl group, a phenylazo group, a phenylureido group, a nicotinamido group, an isonicotinamido group, a picolinamido group, an anilino group and a benzamido group; and
R.sup.2 and R.sup.3 are independently hydrido, C.sub.1 -C.sub.4 hydrocarbyl, hydroxyl, or amino.

R.sup.4 is a phenyl, phenoxy, thiophenoxy, phenylazo, benzamido, anilino, nicotinamido, isonicotinamido, picolinamido or phenylureido group that is optionally substituted:
at the meta- or para-position or both with a moiety that is selected from the group consisting of a halogen, a C.sub.1 -C.sub.9 hydrocarbyloxy group, a C.sub.1 -C.sub.10 hydrocarbyl group, a di-C.sub.1 -C.sub.9 hydrocarbylamino group, a carboxylC.sub.1 -C.sub.8 hydrocarbyl group, a C.sub.1 -C.sub.4 hydrocarbyloxy carbonyl C.sub.1 -C.sub.4 hydrocarbyl group, a C.sub.1 -C.sub.4 hydrocarbyloxycarbonyl C.sub.1 -C.sub.4 hydrocarbyl group and a C.sub.1 -C.sub.8 -hydrocarbyl carboxamido group, or

6. The compound according to claim 4 wherein z is one and y is zero.
7. The compound according to claim 4 wherein y is one and z is zero.
8. A compound according to claim 4 corresponding to Formula II or III: ##STR626##

R.sup.4 is selected from the group consisting of a single-ringed aryl group, a single-ringed heteroaryl group, a phenoxy group, a thiophenoxy group, a 4-thiopyridyl group, a phenylazo group, a phenylureido group, a nicotinamido group, anisonicotinamido group, a picolinamido group, an anilino group, and a benzamido group.
9. A compound corresponding to Formula II or III: ##STR627##
wherein
the ring structure W is a 6-membered divalent aromatic or heteroaromatic ring;
R.sup.1 is a 5- or 6-membered cyclohydrocarbyl, heterocyclo, aryl or heteroaryl radical bonded directly to the depicted SO.sub.2 -group and substituted at its own 4-position when a 6-membered ring or at its own 3- or 4-position when a 5-memberedring with R.sup.4 ;
R.sup.2 and R.sup.3 are independently hydrido, C.sub.1 -C.sub.4 hydrocarbyl, hydroxyl, or amino; and
R.sup.4 is a phenyl, phenoxy, anilino, thiophenoxy, benzamido, nicotinamido, isonicotinamido, picolinamido, or phenylureido group optionally substituted at:

10. The compound according to claim 9 wherein said R.sup.4 is a phenoxy or thiophenoxy group that is unsubstituted.
11. The compound according to claim 8 that corresponds in structure to Formula II.
12. The compound according to claim 8 that corresponds in structure to Formula III.
13. The compound according to claim 8 wherein said 6-membered divalent aromatic or heteroaromatic ring W is selected from the group consisting of a 1,2-phenylene, 2,3-pyridinylene, 3,4-pyridinylene, 4,5-pyridinylene, 2,3-pyrazinylene,4,5-pyrimidinylene, and 5,6-pyridininylene.
14. The compound according to claim 8 corresponding in structure to Formula IIA ##STR628##
wherein PhR.sup.4 is phenyl substituted at its 4-position with R.sup.4.

A process for treating a host mammal having a disease resulting from pathological matrix metalloprotease activity, the process comprising administering a compound in an MMP enzyme-inhibiting effective amount to a mammalian host having such adisease, wherein the compound corresponds in structure to Formula I: ##STR633##

the ring structure W is a 5- or 6-membered divalent aromatic or heteroaromatic ring;
R.sup.1 is a single-ringed cyclohydrocarbyl, heterocyclo, aryl, or heteroaryl that is 5- or 6-membered, and substituted at its own 4-position when a 6-membered ring or at its own 3- or 4-position when a 5-membered ring with R.sup.4 ;
R.sup.2 and R.sup.3 are independently hydrido, C.sub.1 -C.sub.4 hydrocarbyl, hydroxyl, or amino; and
R.sup.4 is selected from the group consisting of a single-ringed aryl group, a single-ringed heteroaryl group, a phenoxy group, a thiophenoxy group, an anilino group, a 4-thiopyridyl group, a phenylazo group, a phenylureido group, a nicotinamidogroup, an isonicotinamido group, a picolinamido group, and a benzamido group.
20. The process according to claim 19 wherein:
R.sup.1 is phenyl substituted at its 4-position with R.sup.4, and
R.sup.4 is a phenyl, phenoxy, anilino, thiophenoxy, phenylazo, benzamido, nicotinamido, isonicotinamido, picolinamido, or phenylureido group.
21. A process for treating a host mammal having a disease resulting from pathological matrix metalloprotease activity, the process comprising administering a compound in an MMP enzyme-inhibiting effective amount to a mammalian host having such adisease, wherein the compound corresponds in structure to Formula I: ##STR634##

the ring structure W is a 5- or 6-membered divalent aromatic or heteroaromatic ring;
R.sup.1 is phenyl substituted with R.sup.4 at the 4-position;
R.sup.2 and R.sup.3 are independently hydrido, C.sub.1 -C.sub.4 hydrocarbyl, hydroxyl, or amino; and
R.sup.4 is a phenyl, phenoxy, anilino, thiophenoxy, phenylazo, benzamido, nicotinamido, isonicotinamido, picolinamido or phenylureido group that is substituted:
at the meta- or para-position or both with a moiety that is selected from the group consisting of a halogen, a C.sub.1 -C.sub.9 hydrocarbyloxy group, a C.sub.1 -C.sub.10 hydrocarbyl group, a di-C.sub.l -C.sub.9 hydrocarbylamino group, a carboxylC.sub.1 -C.sub.8 hydrocarbyl group, a C.sub.1 -C.sub.4 hydrocarbyloxy carbonyl C.sub.1 -C.sub.4 hydrocarbyl group, a C.sub.1 -C.sub.4 hydrocarbyloxycarbonyl C.sub.1 -C.sub.4 hydrocarbyl group and a C.sub.1 -C.sub.8 hydrocarbyl carboxamido group, or

22. The process according to claim 18 wherein R.sup.4 is a phenoxy or thiophenoxy group that is unsubstituted.
23. The process according to claim 18 wherein said compound is administered a plurality of times.
